StratiCELL EPI/001
The StratiCELL® Reconstituted Human Epidermis is produced from human keratinocytes originating from foreskin tissue from a pool of different donors. This unique feature in our manufacturing procedure ensures the highest level of standardization and reproducibility of the test results.
Cells are seeded on polycarbonate filters (standard size 0.63 cm2), grown at the air-liquid interface in an appropriate cell culture medium in an insert (figure 1a). After two weeks of differentiation, a full epidermis is reconstituted.
The StratiCELL® reconstituted epidermis is very similar to human skin. In terms of morphology for instance, the well-characterized layers of a differentiated epidermis, -the basal, spinous, granular and cornified layers (cited from bottom to top)-, are present in the in vitro model as well as observed by a histology assessment (figure 1b). With on the top the appearance of terminally differentiated keratinocytes that form the protective stratum corneum layer. This morphology is accompanied with a highly similar expression profile of different differentiation markers in the reconstituted epidermis in comparison to human skin (such as keratins 10 & 14, involucrin, figure 2). In terms of functional characteristics, the in vitro epidermal model retains, its capacity to synthesize inflammatory cytokines in response to irritating or sensitizing agents.
Each batch of reconstituted epidermis is submitted to quality control (viability and histoloy).
